Amidations are amongst the most common reactions in medicinal chemistry where speed and automation are of particular importance. SiliCycle offers a range of amide reagents for this purpose. A supported reagent, like a scavenger, is a reactive functional group grafted onto insoluble silica. Unlike a scavenger which is added once a reaction is complete, a supported reagent is added at the beginning of the reaction and replaces the solution phase reagent and allows the supported reagent to be used in greater excess, driving the reaction to completion. It also facilitates multiple-step one-pot reactions. The spent reagent is easily removed by filtration. Bound reagents are an excellent alternative in cases where the reagent is used in excess and can be difficult to remove. Purification becomes therefore a simple filtration and evaporation process.
It has been shown that using SiliaBond Carbodiimide and Dichlorotriazine reagents, amides can be synthesized in a matter of minutes, increasing productivity and throughput in the laboratory. |
